Understanding the consequences of the autoimmune attack on the muscle function:
Seronegative MG (SNMG) concerns about 15% of MG patients. These patients do not produce antibodies against AChR. Among them, 30% produce antibodies against a muscle-specific kinase-receptor (MuSK+). The team has shown that anti-Musk antibodies are pathogenic in an in-vitro model [Boneva, 2006]. The pathogenicity is mediated through the effects of the antibodies on Rac/Cdc42 small Rho GTPases that are downregulated after anti-Musk Ab treatment. Since these proteins are involved in AChR clustering and expression, these effects could explain the signs of pathogenicity observed in MuSK+ MG patients.The team is also studying the dysregulation in the expression of genes in the muscle of SNMG and seropositive MG (SPMG) patients by transcriptomics analysis, in order to1) Explore the mechanisms of action of the antibodies and2) Identify new possible targets in SNMG patients.
